Progesterone and Cilostazol Protect mice pancreatic islets from oxidative stress induced by hydrogen peroxide
Authors
Abstract:
Abstract Reactive oxygen species and oxidative stress impair β-cell function and reduce insulin secretion. It has been shown that progesterone and cilostazol possess antioxidant properties. The present study was aimed to investigate in vitro pretreatment effect of progesterone and cilostazol on insulin secretion as well as their protective effects against hydrogen peroxide-induced oxidative stress in pancreatic isolated islets from mouse. Pancreatic islets were isolated from 84 male NMRI mice (25–30g) by collagenase digestion method and pretreated for 48h with cilostazol (10 μM), progesterone (0.5 µM) and glibenclamide (10 μM) in culture medium. Then islets were exposed to hydrogen peroxide (H2O2. 500 μM) for 2h. Next, culture mediums containing glucose concentration of 2.8 mM or 16.7 mM were added to them and incubated in this status for 1h. At the end, the rate of insulin out¬put from islets, lipid peroxidation and antioxidant enzymes activities in islet tissues were assayed. Exposure of islets to H2O2, resulted in a significant decrease in insulin secretion, superoxide dismutase and catalase activities (P

full text8-Alkylcoumarins from the Fruits of Cnidium monnieri Protect against Hydrogen Peroxide Induced Oxidative Stress Damage
Three new 8-alkylcoumarins, 7-O-methylphellodenol-B (1), 7-methoxy-8-(3-methyl- 2,3-epoxy-1-oxobutyl)chromen-2-one (2), and 3'-O-methylvaginol (3), together with seven known compounds (4-10) were isolated from the fruits of Cnidium monnieri. Their structures were determined by detailed analysis of spectroscopic data and comparison with the data of known analogues. All the isolates were evaluate...
full textOxidative stress response of Inonotus obliquus induced by hydrogen peroxide.
While the medicinal fungus Inonotus obliquus produces polyphenols as one of its main metabolites in natural habitats, it accumulates less polyphenols under laboratory conditions. In this study we found that the continuous addition of 1 mM H(2)O(2) at a rate of 1.6 ml/h into a submerged culture of the fungus enhanced its production of mycelia, melanins, flavonoids and hispidin analogs (HA). Simu...
full text12-lipoxygenase promotes obesity-induced oxidative stress in pancreatic islets.
High-fat diets lead to obesity, inflammation, and dysglycemia. 12-Lipoxygenase (12-LO) is activated by high-fat diets and catalyzes the oxygenation of cellular arachidonic acid to form proinflammatory intermediates. We hypothesized that 12-LO in the pancreatic islet is sufficient to cause dysglycemia in the setting of high-fat feeding. To test this, we generated pancreas-specific 12-LO knockout...
